We will present an end-to-end workflow for identifying CD8⁺ T cells in peptide-stimulated PBMCs. DriverMap AIR-TCR sequencing is used to detect TCRs that expand after antigen exposure. Bulk analysis identifies 1–50 enriched TCRs per target, while single-cell AIR-TCR analysis provides TCR chain pairing info. An alternative method for TCR-discovery based on IFNγ secretion on the CloneXplorer platform will be presented. Selected TCRs are cloned and expressed in Jurkat NFAT-GFP reporter cells, and functionally validated by real-time GFP and IFNγ monitoring using the CloneXplorer’s FluoroSpot Assay.
