A standardized workflow was developed to optimize a 22-color immunophenotyping panel for the CytoFLEX LX & Mosaic 88 and applied to a murine house dust mite asthma model. Lung samples from saline controls, acute asthma, and neonatal hyperoxia–exposed mice were analyzed to assess whether early hyperoxia worsens allergic inflammation. The spectral panel enabled high-resolution profiling of key immune populations and activation states, leveraging full spectrum detection to improve subset resolution and characterize how hyperoxia shapes later airway inflammation.
